1. Product description

Sagarose® Ultra-Low Gel Temperature Agarose is a special "soft agarose" product with ultra-low gelling and melting temperature characteristics, making it valuable in a variety of technologies involving biomolecules. Sagarose® Ultra-Low Gel Temperature Agarose is an ideal medium for hybridoma cloning in a soft, crystal clear gel (clarified sol can be stored aseptically at 37 °C for almost indefinitely). Sagarose® ultra-low gel temperature agarose gels below 17 °C and can then melt at 40 °C -55 °C. This extended ultra-low operating temperature range provides great flexibility and convenience for handling heat-sensitive biomolecules.

At concentrations of 0.8%-1.5%, the softgel texture makes Sagarose® ultra-low gel temperature agarose a suitable suspension medium for cells and promotes its recovery by gentle mechanical destruction for subsequent growth in liquid media. Growth factors can be added to Sagarose® ultra-low gel temperature agarose along with other nutrients to prepare "qualified" media.

4. How to use

(1) Agarose autoclaving procedure

1. Choose a flask with a volume of 2-4 times the volume of the solution,

2. Add water to the flask.

3. Sprinkle in the pre-weighed agarose powder at 2× final agarose concentration.

4. Cover the flask with aluminum foil and place the flask in the autoclave.

5. Sterilize at 0.1 Mpa autoclave for 5 min.

Note: Agarose may lose gel strength when exposed to autoclaves for longer periods of time.

6. Once the agarose solution has cooled, aliquot into usable aliquots and store at 4 °C before use.

(2) General method for using agarose in cell culture media

1. Heat the agarose gel in a hot water bath or microwave until completely dissolved.

2. Cool the agarose solution to 37 °C.

3. Preheat the 2× medium solution to 37 °C.

4. Mix an equal volume of sterile 2× agarose solution with an equal volume of sterile 2× medium containing growth factors and nutrients.

5. Pour the agarose/media mixture into a plate or sterile culture tube.

6. If the agarose solution is used as a feeder layer or mulch, let the agarose solution gel for 20 minutes; if it is used as a liquid culture, keep the solution at 37 °C.

5. Precautions

1. This product is only for scientific research use and should not be used in clinical diagnosis and treatment.

2. In applications such as hybridoma cloning, it is recommended to test different batches of ZechinTM low gel temperature agarose to select the appropriate gel strength product. When preparing agarose for cell culture, it should be prepared with water suitable for cell culture and separated from the medium or nutrient solution. The agarose solution and media solution should be prepared at 2 × concentrations (e.g., 1.2% for the target final concentration of 0.6%) and aliquoted after autoclaving separately.
